I’m going to develop a new website in asp. It’ll be an images based website. I want to know if directory structure and filenames matter in ranking. If yes, how much do they matter and what are best practices?

Yes, it does matter. Put your keyword or phrase in your filename, this will give a little push in ranking. The filename should also be no longer then 15-20 characters. If you have vast amounts of data and need to organize it in different directories please add at least one keyword within the name. A directory structure that uses keywords in directory name can additionally benefit.
